Interferon-inducible protein 10, monokine induced by interferon gamma, and interferon-inducible T-cell alpha chemoattractant are produced by thymic epithelial cells and attract T-cell receptor (TCR) ab1CD81 single-positive T cells, TCRgd1 T cells, and natural killer–type cells in human thymus

چکیده
Strong reactivity for interferon-inducible protein 10 (IP-10), monokine induced by interferon gamma (Mig), and interferoninducible T-cell alpha chemoattractant (I-TAC) was found in epithelial cells mainly localized to the medulla of postnatal human thymus. The CXC chemokine receptor common to the 3 chemokines (CXCR3) was also preferentially expressed in medullary areas of the same thymuses and appeared to be a property of 4 distinct populations: CD31T-cell receptor (TCR) ab1CD81 single-positive (SP) T cells, TCRgd1 T cells, natural killer (NK)–type cells, and a small subset of CD31(low)CD41CD81TCRab1 double-positive (DP) T cells. IP-10, Mig, and I-TAC showed chemoattractant activity for TCRab1CD81 SP T cells, TCRgd1 T cells, and NK-type cells, suggesting their role in the migration of different subsets of mature thymocytes during human thymus lymphopoiesis. Expression Pattern of Interferon-γ in Human Leukemic T Cell Lines Following Treatment with Phytoheamagglutinin, phorbol myristate acetate and Lipopolysaccharide
Background: As a T helper type 1 (Th1) derived cytokine, Interferon gamma (IFN-γ) is an important regulator of inflammatory immune responses. Furthermore, IFN-γ plays an essential role in defense against tumors and intracellular pathogens. This study was designed to assess the pattern of IFN-γ production in human leukemic (Jurkat and Molt-4) T cell lines in vitro. Methods: Ju...
متن کاملInterferon–inducible T Cell Alpha Chemoattractant (I-TAC): A Novel Non-ELR CXC Chemokine with Potent Activity on Activated T Cells through Selective High Affinity Binding to CXCR3
Chemokines are essential mediators of normal leukocyte trafficking as well as of leukocyte recruitment during inflammation. We describe here a novel non-ELR CXC chemokine identified through sequence analysis of cDNAs derived from cytokine-activated primary human astrocytes. This novel chemokine, referred to as I-TAC (interferon-inducible T cell alpha chemoattractant), is regulated by interferon...
متن کاملCutting edge: developmental up-regulation of IFN-gamma-inducible lysosomal thiol reductase expression leads to reduced T cell sensitivity and less severe autoimmunity.
Reactivity to self-peptide/MHC complexes is required for selection of the TCR repertoire in the thymus but can also promote autoimmunity. Reduced TCR sensitivity of mature T cells is thought to help control the autoreactivity in peripheral T cells. The molecular basis for reduced sensitivity of peripheral T cells is not known. We found that peripheral T cells, but not immature thymocytes, lacki...
متن کاملP 141: Mesenchymal Stem Cells as Treatment in Neuroinflammatory Disease
Mesenchymal stem cells can be obtained from deferent tissues like adipose tissue, umbilical cord, placenta, skin, bone marrow, etc. These cells have regulatory effects on all types of immune cells such as dendritic cell, natural killers and lymphocytes. Mesenchymal stem cells induce inhibitory phenotypes of Antigen Presenting Cells (APCs) following their activity. They also change T cells pheno...
متن کاملP169: The Role of Lymphocytes in Spinal Cord Injury and Pain; T Helper Cells (TH1 and TH2 Cells)
Lymphocyte is one of the subtypes of white blood cell (WBC) in immune system. Lymphocytes contain T cells, natural killer cells , and B cells. They are the head type of cell found in lymph, which for this reason the name "lymphocyte". Lymphocytes can be recognized by their large nucleus. Infiltration of immune cells in the central nervous system (CNS) helps the start of chronic pain. ...
